Approved uses

Neovascular (wet) age-related macular degeneration

6 mg (0.05 mL of 120 mg/mL) intravitreally monthly for 3 doses, then every 8 to 12 weeks

Carries a labelled risk of retinal vasculitis and vascular occlusion the other anti-VEGF agents do not

Diabetic macular edema

6 mg (0.05 mL of 120 mg/mL) intravitreally every 6 weeks for 5 doses, then every 8 to 12 weeks

Contraindications

Ocular or Periocular Infections Active Intraocular Inflammation Hypersensitivity.

Available Forms

Intravitreal injection, 6 mg (0.05 mL of 120 mg/mL) in a single-dose pre-filled syringe or single-dose vial.

Mechanism of Action

Brolucizumab is a human VEGF inhibitor. Brolucizumab binds to the three major isoforms of VEGF-A (e.g., VEGF 110, VEGF 121, and VEGF 165 ), thereby preventing interaction with receptors VEGFR-1 and VEGFR-2. By inhibiting VEGF-A, brolucizumab suppresses endothelial cell proliferation, neovascularization, and vascular permeability.

Warnings & Precautions

Endophthalmitis and retinal detachment may follow intravitreal injection. Retinal vasculitis and retinal vascular occlusion, typically accompanied by intraocular inflammation, have been reported and may result in severe vision loss; instruct patients to report any change in vision without delay. Increases in intraocular pressure have been seen after injection.

Common Side Effects

The most common adverse reactions reported in patients receiving BEOVU are vision blurred, cataract, conjunctival hemorrhage, eye pain, and vitreous floaters.

Pregnancy & Lactation

Pregnancy: No adequate and well-controlled studies in pregnant women. Intravitreal brolucizumab in pregnant monkeys produced systemic exposures above those expected clinically without a clear effect on the fetus.

Lactation: No data on presence in human milk or effects on the breastfed infant.

Pediatric Use

The safety and efficacy of BEOVU in pediatric patients has not been established.
