Dry eye disease

One drop in each eye twice daily, about 12 hours apart

A TRPM8 agonist: it drives trigeminal signalling to increase basal tear production rather than supplementing tears

Contraindications

None.

Available Forms

Acoltremon ophthalmic solution 0.003% in single-dose vials.

Mechanism of Action

Studies in animals suggest that acoltremon, the active substance in TRYPTYR, is an agonist of transient receptor potential melastatin 8 (TRPM8) thermoreceptors. TRPM8 thermoreceptor stimulation has been shown to activate trigeminal nerve signaling leading to increased basal tear production. The exact mechanism of action for TRYPTYR in dry eye disease is unknown.

Warnings & Precautions

To avoid eye injury and contamination, do not touch the vial tip to the eye or any other surface.

Common Side Effects

The most common adverse reaction was instillation site pain (50%).

Pregnancy & Lactation

Pregnancy: No adequate and well-controlled studies in pregnant women; systemic exposure after ocular use is low.

Lactation: No data on presence in human milk; systemic exposure after ocular use is low.

Pediatric Use

The safety and effectiveness of TRYPTYR have not been established in pediatric patients.
